Chemiluminescent response of polymorphonuclear leukocytes to Streptococcus pneumoniae and Haemophilus influenzae in suspension and adhered to glass.
We measured the luminol- and lucigenin-enhanced chemiluminescent response of human polymorphonuclear leukocytes (PMN) stimulated by various strains of Streptococcus pneumoniae and Haemophilus influenzae. In the absence of opsonin, phagocytosis of either bacterial species elicited good PMN response when the bacteria were adhered to a surface but minimal PMN response when they were in suspension....
متن کاملEffect of phorbol myristate acetate on the oxidative metabolism of human polymorphonuclear leukocytes.
The addition of 0.1 mug/ml of phorbol myristate acetate (PMA) to a suspension of resting human neutrophils causes a marked stimulation of all aspects of cellular oxidative metabolism normally associated with phagocytosis. PMA induces a greatly increased rate of glucose oxidation via the hexose monophosphate shunt, increased production of superoxide anion and of hydrogen peroxide, increased cel...
متن کاملStaphylococcal protein Ecb impairs complement receptor-1 mediated recognition of opsonized bacteria
Staphyloccus aureus is a major human pathogen leading frequently to sepsis and soft tissue infections with abscesses. Multiple virulence factors including several immune modulating molecules contribute to its survival in the host. When S. aureus invades the human body, one of the first line defenses is the complement system, which opsonizes the bacteria with C3b and attract neutrophils by relea...
متن کاملHuman peritoneal macrophage phagocytic, killing, and chemiluminescent responses to opsonized Listeria monocytogenes.
Opsonization with normal human serum, purified immunoglobulin G, or immunoglobulin G-deficient serum promoted phagocytosis of Listeria monocytogenes by human peritoneal macrophages. However, normal human serum was the most effective opsonin in elicting killing and chemiluminescent responses. Macrophages phagocytized and killed almost as much as polymorphonuclear leukocytes but produced consider...
متن کاملRegulation of human monocyte/macrophage function by extracellular matrix. Adherence of monocytes to collagen matrices enhances phagocytosis of opsonized bacteria by activation of complement receptors and enhancement of Fc receptor function.
In inflammation monocytes emigrate from the peripheral circulation into an extravascular area rich in extracellular matrix proteins. In this milieu, phagocytes ingest and kill invading pathogens. In the present studies, we found that monocytes adhered to type I collagen gels phagocytized 2.5-12-fold more opsonized Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes, and Streptococcu...
